Spectroscopic feature detection for age determination of human blood samples in the laboratory and at the crime scene

  • In der Forensik sind Blutspurenanalysen ein wichtiges Mittel der Tatortermittlung. Aus diesem Grund wäre die Altersbestimmung von Blut ein guter Ansatz, um die Aufklärungsrate noch zu verbessern. In dieser Arbeit mit Fokus auf Grundlagenforschung wurden durch spektrophotometrische Analysen und statistischer Auswertung zwei große Komplexe untersucht. Vergleichende Analysen von Schweineblut mit Humanblut wurden durchgeführt und geprüft, ob sie in ihren Ergebnissen vergleichbar sind. So sollten Rückschlüsse auf die Übersetzung bereits vorhandener Studien mit Schweineblut ermöglicht werden, um die Eignung als Humanblutersatz zu diskutieren. Weiterhin wurde durch Vergleich der Absorptionsspektren über Zeit ermittelt, wie stark und auf welche Weise die Zugabe von Gerinnungshemmern wie Citrat Einfluss auf den Prozess der Blutalterung nimmt. In einem zweiten Komplex wurde versucht, eine auf Spektro-Photometrie basierende statistische Methode zu etablieren, die den Prozess der Blutalterung nachvollziehen und mit möglichst geringen Abweichungen vorhersagen kann. Dies sollte potentiell die Bestimmung des Alters einer Blutprobe ermöglichen. Im Zuge dessen wurden eine Hypothese zu den ex vivo ablaufenden chemischen Prozessen der Blutalterung erörtert und anhand der Spektraldaten diskutiert. Charakterisiert wurde dies über die messbaren Anteile der Hämoglobin-Derivate im UV-Vis-Spektrum einer Vollblutprobe
  • In Forensics, blood stain analysis is one of the most important indication of crime (scene) investigation. So age determination of blood samples would be a good attempt to enforce the enlightment rate of crimes. In this thesis, which is focused on basic research, two large parts were examined using spectro-photometric and statistical analysis. Comparative analyses of pig's blood with human blood have been used to find out, if they are comparable in their results. Therefore conclusions should be made to examine if existing studies of pig's blood could be transfered to human blood in order to discuss suitability as a human blood substitute. Furthermore, by comparing the absorption spectra over time, it was determined how strongly and in what way the addition of anticoagulants, such as citrate influences the process of blood aging. In a second part, an attempt was made to establish a statistical method based on spectro-photometry, that can reflect the process of blood aging ex vivo and predict it with as little variation as possible. This should potentially allow to determine the age of blood stains. Furthermore, a hypothesis on the ex vivo chemical processes of blood aging, based on research and spectral data, was discussed. This was characterized by the measurable proportions of hemoglobin derivatives in the UV-Vis spectrum of a (whole) full blood sample.
