  • DNA Analysen werden immer wichtiger, wenn es um die Aufklärung früherer Lebens- und Familienverhältnisse geht. Viele DNA Proben sind durch verschiedene Umwelteinflüsse degradiert und die DNA ist meist weniger als 300 bp lang. Eine Analyse solcher Proben ist kompliziert. Um erfolgreiche Analysen für forensische und anthropologische Zwecke zu gewährleisten, ist die Entwicklung neuer Methoden wichtig. Die Firma Promega hat in diesem Jahr ein neues DNA Quantifizierungskit auf den Markt gebracht, welches sich PowerQuant TM System nennt. Ziel dieser Arbeit ist es zu testen, ob sich das Kit zur Analyse degradierter DNA Proben eignet. Außerdem soll getestet werden, wie genau das Kit Inhibitoren detektieren kann, die sich eventuell in der Probe befinden. Das historische Knochenmaterial, welches zur Analyse verwendet wird, stammt dabei von Gräberfeldern aus Görzig und Dresden-Briesnitz.
  • DNA analysis is very important to find out more about the family and lifestyle of people who lived in the past. But many DNA samples are degraded due to environmental influence and in a lot of cases, the DNA is only less than 300 bp long. The analysis of such samples is difficult. For the successful application of commercial DNA quantification systems for ancient DNA (aDNA) analyzes for forensic and anthropological purpose, the development of appropriate sensitive tools is necessary. A new generation of DNA quantification kit called PowerQuant TM was provided by Promega this year. Purpose of this work is to see if this Kit is suitable to analyze aDNA samples with regard to highly degraded DNA and its sensitivity to display the presence of possible PCR inhibitors. The sample batch consists of skeletal material from burial ground Görzig and Dresden-Briesnitz.
