Entwicklung von Assays zur Detektion von niedermolekularen Substanzen mit Aptameren am Beispiel von 17ß-Estradiol

  • Die ubiquitäre, anthropogene Ausbreitung von pharmazeutischen Substanzen in der Umwelt, stellt ein weitreichendes Problem für alle Lebewesen dar. Hormone, die auf unterschiedlichsten Wegen in den aquatischen Lebensraumgelangen, sind unter anderem verantwortlich für das Fischsterben. Da sich die Anwesenheit dieser Stoffe allgemein und besonders bei Hormonen in der Umwelt nur auf geringe Konzentrationen begrenzt und sie dabei dennoch ein großes Risiko darstellen, sind geeignete Methoden zum Nachweis und zur Entfernung dieser Stoffe aus dem aquatischen System notwendig. Diezbezüglich sollen bereits etablierte biosensorische Detektionsmethoden unter Einsatz von Aptameren untersucht werden, um niedermolekulare Substanzen zu detektieren. Das Zielmolekül stellt dabei das Hormon 17ß-Estradiol, dass auf der Beobachtungsliste der EU Wasserrahmenrichtlinie steht und zu welchem bereits spezifische Aptamere veröffentlich sind, dar.
  • The ubiquitous, anthropogenic distribution of pharmaceutical substances in the environment is amajor problem for all living organisms. Hormones entering the aquatic ecosystem can, among other things, account for fish mortality. Pharmaceuticals in generally and specifically hormones occur often only at low concentrations in the environment but still pose a tremendous threat. Therefore it will be important to develop effective technologies to reduce hormone concentration in the aquatic system and suitable methods for the detection of these substances are necessary. Toward the goal of detecting small molecules established biosensor methods will be utilized and the use of aptamers will be investigated for the detection of the target molecule 17ß-Estradiol. This hormone is included in the observation list of the EU Water Frame Directive and specific aptamers are already published.
